چکیده

Abstract Background & Purpose: Gut acute graft-versus-host disease (aGVHD) is a serious complication after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and associated with high mortality. Mucosal-associated invariant T cells (MAITs) are group of innate-like enriched in the intestine that can be activated by riboflavin metabolites from various microorganisms. However, little known about function or mechanism action MAIT occurrence gut aGVHD human body. Methods Cases: In our study, multiparameter flow cytometry (FCM) was used to evaluate number functional cytokines. 16S V34 region amplicon sequencing analysis analyse intestinal flora transplant patients. vitro stimulation coculture assays were study activation MAITs. The distribution MAITs tissues analysed immunofluorescence technology. We prospectively studied enrolled 150 consecutive patients who underwent allo-HSCT institute. Results: frequency infused grafts lower than those non-gut (Figure). Recipients had higher abundance early post-transplantation period (+14 days). At onset aGVHD, decreased peripheral blood, marker CD69, chemokines CXCR3 CXCR4 transcription factors Rorγt T-bet tended increase. Furthermore, when occurred, proportion MAIT17 MAIT1. non-riboflavin metabolic pathways increase secreted more granzyme B, TNF-α IFN-γ under IL-12/IL-18 (non-TCR signal) most IL-17 CD3/CD28 (TCR signal). inhibited proliferation CD4+ vitro. Conclusions: conclusion, related incidence may affect composition recipients transplantation. metabolism pathway promoted secretion protective humans. Figure 1 1. Disclosures No relevant conflicts interest declare.

Relationship between Molecular Chimerism and Graft Versus Host Disease after Allogenic Hematopoietic Stem Cell Transplantation

Background: Bone marrow transplantation (BMT) is considered as a curative therapy for a broad range of diseases. However, complications such as relapse and graft versus host disease (GVHD) may be observed following BMT. Chimerism analysis serves as a reliable indicator of transplant outcome. Complete chimerism refers to the complete replacement of hematopoietic system by donor cells, while mixe...

Assessment of Cyclosporine Serum Concentrations on the Incidence of Acute Graft versus Host Disease Post Hematopoietic Stem Cell Transplantation

Allogeneic hematopoietic stem cell transplantation (HSCT) is a curative treatment option for hematological disorders. Cyclosporine (CsA) is one of the major immunosuppressive agents for the prophylaxis against graft versus host disease (GvHD). In this retrospective study, we evaluated the effects of CsA serum levels on the incidence of acute GvHD and transplant outcomes. 103 adult patients rece...

Invariant Natural Killer T Cells As Suppressors of Graft-versus-Host Disease in Allogeneic Hematopoietic Stem Cell Transplantation

Invariant natural killer T (iNKT) cells serve as a bridge between innate and adaptive immunity and have been shown to play an important role in immune regulation, defense against pathogens, and cancer immunity. Recent data also suggest that this compartment of the immune system plays a significant role in reducing graft-versus-host disease (GVHD) in the setting of allogeneic hematopoietic stem ...

tgf-β codon 25 polymorphism and the risk of graft-versus-host disease after allogenic hematopoietic stem cell transplantation

some of the genotypes of cytokines are associated with acute graft versus host disease after bone marrow transplantation. the purpose of the present investigation was to find out the possible association between transforming growth factor beta-1 (tgf-β1) codon 25 polymorphism (rs:1800471) and acute graft versus host disease (agvhd) after bone marrow transplantation from the sibling with the sim...
